midlife single women

Hi,
I'm 45, single, never married and childless. I don't think I regret not having kids. I have given up on ever marrying.
My life for the past year and a half has been sheer mental torture, trying to decide if I should quit my job. I never really liked my field of work to begin with (business admin), but my job became my life and my status was wrapped up in titles and promotions. Now I have no friends to speak of and everyone is busy with their own lives. I've read that people should not make radical changes at midlife, but I am wondering why not? Why continue in misery if I am only responsible for myself? I actually feel a calling to the religious life. Has anyone experienced that? And what did you do about it?
